While Vibrational Circular Dichroism (VCD) spectra carry a great deal of information about chiral molecules, the experimental implementation of this method to proteins and peptides has been limited by very weak signals and the need for photoelastic modulators in the mid-IR. We aim to show that measurement of vibrational optical activity using a simple attenuated total reflectance (ATR) setup, and an IR quantum cascade laser (QCL), for dry and membrane bound peptides, is achievable with adequate signal-to-noise and all the benefits of reflection spectroscopy.
