Guanine-quadraplex (GQ) is an unusual secondary DNA structure formed by guanine quartets associated together by Hoogsteen hydrogen bonds.  Recent studies have revealed GQ presence in telomeres and oncogenes, sparking biomedical interest.  The interaction of small molecule ligands with GQ DNA has been shown to lead to tumor suppression; stabilization of telomeric DNA by ligands may inhibit telomerase, the protein responsible for telomere maintenance in 85% of tumors.  The GQ field is in search for efficient and selective GQ stabilizers.  Here, we present the results of our evaluation of two categories of promising metallic complexes, platinum metalloporphyrins and octahedral ruthenium complexes.  The latter contain two bipyridine ligands and one dipyridophenazine or pterinylphenanthroline ligand.   Fluorescence resonance energy melting studies (FRET), circular dichroism (CD) spectroscopy, and UV-Vis titrations were employed to assess the properties of these complexes.
