The effect of individual and combined polymers (EudragitE100 and PVPK90) on indomethacin precipitation in solution and their solubility enhancement mechanism from solid dispersion using 1H NMR was studied. The solution studies were done in 0.01N HCl at supersturation level of 3.17. The precipitate was characterized by Raman and SEM. Binary/ternary solid dispersions were prepared and characterized using XRPD, DSC, and Raman. The solubility was assessed in 0.01N HCl. Solution 1H NMR was done to study the site of interaction between drug and polymer. A synergistic precipitation inhibition effect was observed compared to individual polymer. The precipitate showed different morphology in presence of single or combined polymers. The enhancement in solubility was observed from solid dispersion. The difference in the level of supersaturation maintenance and morphology was due to the different site of interaction of PVP K90 and Eudragit E100 with indomethacin as evident from 1H NMR.
