Chemical immobilization of peptides on surfaces is very important in many applications such as antimicrobial coatings and biosensors. The conformation and orientation of such immobilized peptides determine the functions and activities of these peptides. Sum frequency generation (SFG) vibrational spectroscopy was applied to investigate molecular structures of immobilized peptides on self-assembled monolayer (SAM) surfaces. Peptides with different secondary structures such as alpha- helices and beta-sheets have been included in this study. Chemical immobilizations of peptides via different amino acids such as those on the C-terminus or N-terminus, or in the middle of the peptides have been used. The structural changes of these immobilized peptides in dry air, in water, and in air with different humidity levels were also investigated.
