We have developed a sequence-specific fluorogenic �click� reaction that is capable of detecting DNA single nucleotide polymorphism (SNP). A non-fluorescent alkyne-modified coumarine has been successfully incorporated into DNA. The fluorogenic click reaction occurred between azide- and alkyne-modified DNA probes in the presence of matched DNA-template, which produced strong fluorescence with the formation of a triazolyl coumarin. The Fluorogenic click reaction has been used for the detection of the tumor's oligonucleotide. Only the existence of the tumor's oligonucleotide led to the formation of fluorescent triazole complex at room temperature.
