A motivation of our research is to make inroads in the daunting task of chemical analysis related to both complexity and insolubility. Ion mobility spectrometry-mass spectrometry is a crucial aspect of this effort as is new approaches to ionization. Developments in matrix and solvent assisted ionization free of laser or voltage and having advantages of operation at atmospheric pressure or vacuum, speed of analysis, high spatial resolution for imaging, mass range extension, advanced fragmentation capability, and improved ion mobility separation common with multiply charged ions will be presented.  Using this suite of new ionization methods, we have compared structures of peptides and small proteins ionized directly from the solid state, even the native environment of mouse brain tissue, with structures observed from solution using ESI.  We show that these new ionization methods, both AP and vacuum, produce ions of similar structures as determined by IMS-MS (SYNAPT G2) drift time comparisons.
