The Appella lab has developed a DNA/peptide nucleic acid(PNA)-based scaffold that provides increased activity of ligands that specifically target biological sites.  The display of these ligands on our PNA scaffold allows for easily modifiable and specific local concentrations of the ligands. I have synthesized several small molecule ligands, including a cyclic-RGD-mimic to demonstrate the versatility of this scaffold in molecular targeting.  A Gd-chelate was also designed and synthesized as a potential imaging tag to follow the location and concentration of the PNA scaffold. The design and results from investigating the multivalent display of these ligands will be presented.
