We present a brief summary of scanning probe microscopy probing studies of various synthetic and biological macromolecular materials and structures which include  surface force spectroscopy, colloidal force spectroscopy, and high-resolution nanomechanical micromapping.  Data processing is conducted by applying Sneddon's model, layered model, and Johnson viscoelastic model for different cases.  Examples of nanoprobing of gel LbL microcapsules in dry and swollen states, LbL films, segregated polymer brushes, biohydrogel and silk films are presented and discussed.
