Fucosylation has been identified as an important type of post-translational modification in many development processes, such as neural development. Reports have shown that disruption in fucosylation metabolic pathways, resulting in the loss of fucose on the cell surface, brings about severe nerve system development deficiency in zebrafish. However, it has been not clear how fucosylation leads to the neural deficit due to the participation of multiple enzymes and at multiple possible fucosylation sites. In this work, we synthesize a series of GDP-fucose analogues using a chemoenzymatic method and tested their biological effects in vivo by injecting into both wild type and towhead (twd) zebrafish mutant embryos, in which vagus motor neuron progenitors stray medially away from the normal migratory pathway.
