The green fluorescent protein (GFP) and its family have been emerging as powerful probes for intracellular dynamics.1 The avGFP from the jellyfish Aequorea victoria is a globular protein of 238 amino acids folded into 11-stranded �-barrel. At the center of the �-barrel sits a p-hydroxybenzylideneimidazolinone (p-HBI) chromophore.1 The avGFP shows intermolecular excited-state proton transfer (ESPT).1 When the 11-stranded �-barrel is removed from the avGFP, the central free chromophore of p-HBDI (dimethyl derivative of p-HBI) does not display intermolecular ESPT in all solvents. We wondered how replacement of hydroxyl group of p- or o-HBDI with amino functional groups might alter its photo-physics.This poster will show photo-physics of o-(N,N-dimethylamino), o-amino, p-(N,N-dimethylamino) and p-amino analogues of GFP.
