Carolacton, a 12-membered macrolide ketocarbonic acid, was isolated from S. cellulosum Sp ce960 in 1998. More recently, it has been shown to specifically target cells within a biofilm produced by S. mutans with an IC50 value of 15 nM while not affecting planktonic cells. A concise, efficient, and diversity-oriented approach toward the total synthesis of carolacton and a library of analogs will be presented.
