GSNO is an important intermediate in nitric oxide metabolism and mediates many NO-signaling pathways. In the current study, we describe the utilization of LC-MS/MS for the detection and quantitation of GSNO in cancer cells by selectively capturing and converting unstable GSNO to a stable product using a triphenylphosphine-thioester. Importantly we demonstrate that, product ion m/z 309 derived from the phosphine-GSNO adduct can be used to probe the mobile pool of NO and to identify other LMW-SNOs in the cell lysate.
