A� peptide is the primary causative agent of Alzheimer's Disease and its interaction with Cu(II) is believed to be a key event underlying this disorder. The exact role of the A�:Cu(II) interaction is not fully understood but direct binding of Cu(II) to A� leads to generation of neurotoxic A� oligomers and ROS. Many studies focused on quantifying the Cu(II) affinity to A�. Depending on the analytical techniques used and peptide concentration reported values of Kd for a monomeric Cu(II)-A� complex span a range of more than two orders of magnitude (from 5 � 107 to 104  M-1). Our studies aim at designing a competitive strategy with the use of novel peptides to evaluate the method and concentration independent stability of the A�-Cu(II) complex. We also used potentiometric titrations in noncanonical concentrations. Our results support the existence of Kdapp dependence on concentration. This finding is discussed in terms of alternative stoichiometry interactions.
