This presentation will discuss challenges relevant to CSF or plasma protein biomarker discovery and verification using mass spectrometry based platforms. Advances in the quality, resolution, and the speed of separations have arguably been as important as mass spectrometric developments in improving the sensitivity and coverage of proteomics measurements.  However, significant challenges remain related to: the detection of very low abundance proteins, sample size limitations, the need for sufficient throughput to address large numbers of samples, and the ability to effectively distinguish protein isoforms and utilize protein modifications. This presentation will describe new developments involving advanced separations, including multidimensional nanoscale LC and/or ion mobility separations, and the application of improved targeted measurements, and illustrate the potential for addressing these challenges in the context of several CSF and plasma biomarker discovery and verification efforts.
