To develop safety screening commensurate with the level of nanotechnology growth requires high throughput approaches and a predictive toxicological paradigm that links nanomaterial properties to in vitro toxicological outcomes that are also relevant to in vivo health effects. This talk will delineate the use of a multi-parametric high throughput screening assay for toxic oxidative stress to demonstrate its ability to predict the acute inflammatory effects of 24 oxide nanoparticles in the lung based on semiconductor properties, particle dissolution and the ability to generate reactive oxygen species based on band gap overlap with cellular redox potential. I will demonstrate the use of in silico and statistical tools to establish in vitro hazard ranking and structure-activity relationships that can be used for in vivo hazard ranking. I will also provide examples of other predictive toxicological paradigms emerging from the UCLA Center for NanoBiology and Predictive toxicology.
