Development of highly efficient metal-catalyzed cross-coupling reactions remains an important subject in synthetic organic chemistry. Several keyissues include tolerance of steric hindrance, compatibility of various functional groups, and high control of stereochemistry.To address these difficult issues, we seek to develop an efficient monophosphorus ligand to enhance both the reactivity and selectivity of its metal catalyst. In this talk, the syntheses of tetra-ortho-substituted biaryls by a sterically demanding Suzuki-Miyaura coupling will be discussed and constructions of chiral biaryls via asymmetric catalytic Suzuki-Miyaura coupling will be presented. Synthetic efforts towards several chiral biaryl natural products will be disclosed.
