Biodegradable scaffolds for vascular reconstruction have been studied in these decades, but intimal thickening and the occlusion are still their big problems.  We are then challenging to reconstruct native vascular tissues by use of a biodegradable guide which leads to the intimal reconstruction at its luminal surface.  The �vascular tissue reconstruction guide� was made of artificial polypeptides, which were expressed in the E.coli culture. Tubular structure of the hydrogel (i.d.=1mm) was prepared from the mold and covered with the PLLA nanofibers. The guides were transplanted to the rat abdominal aorta, and blood flow was confirmed by MRI and laser-Doppler analysis until 1 month. The vascular tissue structure with smooth muscle cells and endothelial cell was generated in the lumen and showed good patency. These results suggested that the small-caliber blood vessels were reconstructed in the vascular tissue reconstruction guides by the GTR (guided tissue regeneration) mechanism.
