Capillary isotachophoresis (cITP) coupled to microcoil NMR was used to separate and characterize trace amounts of heparin-derived oligosaccharides. The new anionic cITP buffer system featuring a tricine trailing electrolyte buffer significantly lowered the electrophoretic currents produced during the experiments, reducing the effects of current induced broadening on the NMR spectra acquired.  In addition to focusing all 12 commercially-available heparin disaccharides and the Arixtra pentasaccharide, cITP-NMR spectra were measured for components in the complex mixtures of hexasaccharides from heparin, heparan sulfate, and enoxaparin. The results obtained demonstrate the potential of anionic cITP-NMR for evaluating sample purity, composition, and reaction processes for mass-limited heparin and heparan sulfate samples.
