Ewing's sarcoma is a cancerous bone tumor that affects children and young adults. The tumor often metastasizes to the lungs and other bones. Treatment options for patients with Ewing's sarcoma remain unsatisfactory. Our research has focused on the ability of the synthetic compound ajulemic acid to induce death in cultured Ewing's sarcoma cells as well as in a bioluminescent mouse model. Our data show ajulemic acid kills Ewing's sarcoma cells as well as several other pediatric tumor cell lines in vitro, and we have seen complete tumor regression in approximately 33% of our treated mice. The drug also inhibits angiogenesis; this should help limit tumor size and decrease metastases. Thrombospondin is a secreted protein with antiangiogenic effects. In our study, various cell lines were treated with ajulemic acid and thrombospondin concentrations were determined. Our goal is to see if modulation of thrombospondin is the mechanism AJA is using to inhibit angiogenesis.
