The design of a stereoselective approach for the construction of azabicyclic compounds and application towards the total synthesis of several alkaloid natural products will be described. These syntheses feature the use of tert-butyl-sulfinamide as a chiral directing group to form the desired azabicyclic ring moiety in excellent yield and high enantiomeric excess. This highly convergent route has enabled structure activity relationship studies of the azabicyclic natural products at the delta opioid receptor, a therapeutic target for nociception and depression.
