Acetate accumulation during industrial fermentation has a profound effect on the productivity of Escherichia coli, a commonly used model organism for the production of biofuels, proteins, and other fine chemicals. This sensitivity necessitates complex glucose feeding schemes to reduce E. coli acetate production or metabolic engineering to eliminate acetate excretion. An innately acetate-resistant strain of E. coli would therefore be highly desirable for reducing overall process costs and improving specific productivity. To that end, we evolved 12 E. coli populations using serial batch culture in glucose minimal medium supplemented with acetate for 140 generations. Likely acetate resistance mechanisms identified using phenotypic screening and genome sequencing of the isolates are discussed, along with the potential for rational engineering of acetate tolerant industrial strains.
