We describe the synthesis and evaluation of a series of new fluorescent probes (LysoProbe I-VI ) that facilitate monitoring of intracellular pH and labeling of acidic organelles. As stable and sensitive markers for lysosomes, LysoProbes I - VI  have distinct advantages over commercially available lysosomal markers because their fluorescent signals are highly stable. Accordingly, LysoProbes I-VI  provide new tools for monitoring acidic intracellular pH and for the characterization of lysosomal morphology and trafficking in intact cells. We predict that LysoProbe will enable lysosomal labeling of cancer cells and solid tumor tissues, an important step in monitoring the role of lysosomal trafficking in cancer invasion and metastasis.
