Cardiovascular disease (CVD) results in over 810,000 American deaths with estimated costs exceeding three-hundred billion dollars annually. Research indicates that oxidation of low-density lipoprotein (LDL) may initiate atherosclerotic CVD. Thiobarbituric acid reactive substances assay (TBARS) is a widely used method for assessing lipid peroxidation of LDL; however, the results are hard to compare due to variations in protocols used between laboratories. Copper, hypochlorite, and superoxide mediated oxidation of LDL in our lab revealed that low micromolar concentrations of copper where sufficient to generate large TBARS signals while superoxide and hypochlorite require much higher doses. Copper and superoxide dosing showed a linear dose response while hypochlorite required millimolar concentrations to generate a TBARS signal and quickly reached a maximum of lipid peroxide products formed. Further work is in progress to determine the effects of using combinations of copper, hypochlorite, and superoxide.
