In this work, a new boronate affinity chromatographic material, poly(NIPAAm-AAPBA) grafted silica featuring thermally switchable property, was prepared and then used for the capture and release of cis-diol biomolecules through simpler regulation of temperature under physiological pH without changing the composition of mobile phase. The material was designed and prepared by surface-initiated atom transfer radical copolymerization of NIPAAm and AAPBA on silica surfaces. The temperature-controlled method was successfully employed for the enrichment of cis-containing compound, adenosine from the mixture with desoxyadenossine. It was found that the deoxyadenosine was not retarded at various temperatures, while the retention time of adenossine was increased remarkably with the decreasing temperature. A critical change in retention time was observed at around 30?. At the temperatures below 30?, the copolymer chains are extended and the AAPBA is exposed. In this case, cis-diol-compounds are captured. By contrast, the chains become shrinking and AAPBA was shielded at the temperatures above the LCST. In this case, the captured adenossine was released. The selective capture and release was achieved only by change the temperature, temperature seems to act as an on-off signal for the capture-release of the adenosine. These obeservations may provide us with an excellent prospect for the enrichment of glycoproteins at a pH close to physiological pH.
