We have developed various nucleic acid-based lateral flow biosensors (NALFB) for point-of-care biomedical and environmental applications. The NALFBs were fabricated with DNA/aptamer probes, DNA probe-functionalized gold nanoparticles (Au-NP) and lateral flow device. Combining the unique optical properties of Au-NPs and recognition characteristics of DNA probes, the NALFBs have been used for visual detection of specific nucleic acid sequences, Single-nucleotide polymorphism, protein biomarkers, cancer cells and heavy metal ions without instrumentation. The quantitative detection was performed by reading the intensities of the produced red bands with a portable strip reader. The NALFBs thus provide a rapid, sensitive, low cost and quantitative tool for the detection of biological /environmental samples. It shows great promise for in-field and point-of-care diagnosis of genetic diseases, detection of infectious agents, warning against bio-warfare agents or environmental monitoring.
