Protein-protein interactions are essential to current disease and viral detection methods. In this research, we establish nanoparticle tracking analysis (NTA) as a viable method to study these interactions and relevance for low level antigen detection.  Bioconjugation of gold nanoparticles (AuNP) served as the platform for this study. Protein A was adsorbed onto AuNPs and a measurable size increase was observed. Binding affinity of protein A to IgG was also observed.  Additionally, an assay for mouse IgG detection was developed using NTA to detect binding to antibody conjugated AuNPs. The inverse of this AuNP conjugated system, in which IgG conjugated AuNPs are used to detect antibody concentration, was used to measure the binding affinity. NTA results attained were validated by comparison with dynamic light scattering (DLS).  Herein, NTA can be instituted as an analytical tool for AuNP bioconjugation, studying protein-protein binding, and bioassays for low level detection of antigen.
