Microdialysis sampling is a diffusion-based separation in which perfusion fluid passes through a semi-permeable membrane allowing solute collection from complex matrices including tissues. One significant challenge to microdialysis is recovering proteins that may have strong non-specific adsorption to membrane materials. Two proteins, interferon-? (IFN-?) and neuropeptide Y (NPY) exhibit poor relative recovery during in vitro microdialysis sampling. In this work, various perfusion additives were tested to improve recovery. Interestingly, altering bovine serum albumin (BSA) to the perfusion fluids in ranges of 1 to 6% (v/v) did not alter recovery of IFN-?. Other proteins used to block non-specific adsorption are being tested as additives to perfusion fluids. In addition to perfusion additives, the development of ligand-binding assays is ongoing to improve recovery. Particular attention is being paid to the on/off kinetics between proteins and their ligands using surface plasmon resonance (SPR) techniques as a means to predict recovery improvement.
