Studies of an optimized methodology have been explored for the analysis methamphetamine with the use of isotopic dilutions of multiple deuterated methamphetamine analogs that are analyzed by LC-MS TOF.  Methamphetamine D5, D8, D11 and D14 at amounts of 100 ng, 500 ng, 1000 ng and 2000 ng, respectively are added to an unknown sample at the beginning of the extraction sequence. The approach of using multiple deuterated analogs of methamphetamine at different concentrations makes it possible to establish a linear calibration curve from 100 � 2000 ng/mL with a single injection. This technique eliminates matrix effects because the sample itself becomes the matrix. In addition, the extract can be analyzed directly, eliminating the need of time consuming derivatization that is commonly used in traditional drug analysis by GC-MS.  The concentration of methamphetamine in the sample is determined from the calibration curve established by the different concentrations of the deuterated analogs.  It is proven that the mass spectrometer response to methamphetamine-D0 is identical to the response to its deuterated analogs, therefore supporting the integrity of this new methodology.
