Multicomponent reaction chemistry (MCR) brings together several starting material classes in one chemical step and in a convenient one-pot fashion. Many MCRs yield a priory heterocycles and other heterocycles are accessible by the use of bifunctional orthogonal starting materials and a subsequent cyclisation reaction. This concept is very powerful as it allows for the efficient and versatile synthesis of myriads of heterocyclic scaffolds. Recent progress of heterocyclic synthesis using MCR has also lead to several compounds undergoing clinical studies for the treatment of unmet medical needs. Khoury, K., Sinha, M.
