An increasing interest has arisen in characterizing protein interactions of high-concentration solutions, as they provide means to predict and control static and dynamic properties of protein solutions. Classically, virial expansions and simplified models have been combined with Rayleigh scattering data to characterize colloidal interactions in the high-concentration regime. However, such approaches are limited by model approximations and/or the effective concentration range. Here, a novel approach is presented to evaluate concentration dependant colloidal interactions from light scattering (LS) data via the protein-protein Kirkwood-Buff integral. This model is compared against existing approximations for simulated LS data, as well as experimental data for insulin and gamma-D crystallin. The results illustrating this approach provides quantitatively accurate values for protein interactions vs. concentration from low to high concentrations without assumptions on the nature of interactions and/or the thermodynamics of the system.
