Exogenous contrast agents are administered to patients in approximately half of all magnetic resonance imaging (MRI) clinical scans. Paramagnetic Chemical Exchange Saturation Transfer (paraCEST) agents produce contrast based on exchange of labile macrocycle protons (�NH or �OH) with bulk water protons, following saturation of nuclear spins at a particular radio frequency in which contrast is turned on. Paramagnetic high spin iron(II) azamacrocycle complexes based on TACN and Cyclen are a promising addition to the repertoire of trivalent lanthanide paraCEST agents currently under development for clinical use as temperature- and pH-responsive probes. Thermal ablation therapies may benefit from introduction of paraCEST contrast agents that contain narrow exchangeable proton signals. Different complexes exhibit varied responses to pH in the physiologically relevant range, making these complexes promising toward the development of paraCEST MRI contrast agents which may improve diagnosis and prognosis of diseases such as cancer by monitoring extracellular tumor pH.
