DNA lesions are generated by chemotherapeutic drugs, such as platinum drugs, environmental chemical agents, as well as metabolic byproducts from cells. These DNA lesions will interfere with RNA polymerase II (pol II) transcription, if not repaired.  Understanding the functional interplay between the DNA lesions and pol II transcription machinery are essential not only for us to understand the conseuqences of DNA modifications on transcription process but also help us to elucidate the mechanisms of action and resistances of chemotherapeutic drugs. We will discuss our recent progresses toward understanding the functional interplay between DNA damage and transcription machinery.
