Polypeptoids are a class of peptidomimetic polymers featuring N-substituted polyglycine backbones with proteinogenic or synthetic side chains on the nitrogen atoms. They are potentially useful for various biomedical and biotechnological applications due to their enhanced stability against proteolysis, biocompatibility and thermo-processability. The polypeptoid properties such as solubility, crystallinity and conformation are strongly dependent on the side chain structures. Cyclic or linear polypeptoids with controlled chain length and narrow chain length distribution can be synthesized from organo-mediated polymerization of N-substituted N-carboxyanhydride monomers using N-heterocyclic carbene or primary amine initiators. The solution behavior of cyclic and linear polypeptoids (e.g., cloud point transition, gelation) will be discussed with a focus on architectural effect on the respective solution property.
