Free radical oxidation of polyunsaturated fatty acid residues of membrane phosholipids generates hydroperoxides and cyclic peroxides. These initial oxidation products are converted by metal-dependent and -independent reactions to a range of derivatives some of which are reactive electrophiles. Lipid electrophiles react with DNA and protein to block DNA replication, induce mutations and alter cell signaling  inter alia. Comprehensive profiling of the DNA adducts and protein targets of these electrophiles coupled to the cellular responses they induce allows the construction of a reactive electrophile response network. The construction of this network will be described and its use in understanding specific cellular responses to electrophile generation will be illustrated.
