Since current treatments of cyanide poisoning have key disadvantages, a novel antidote, 3-mercaptopyruvate (3-MP), is being investigated. The development of this therapeutic requires an analytical method to detect 3-MP in biological fluids. Therefore, a liquid chromatography-tandem mass-spectrometry (LC-MS-MS) method was established to analyze 3-MP from plasma. An internal standard was added to the samples, proteins were precipitated, the samples were centrifuged, and the supernatant was collected. The sample was dried and modified with monobromobimane (MBB). The 3-MP-MBB complex was separated using a reverse phase column to produce a limit of detection of 100 nM with a linear dynamic range of 200 nM to 100 �M. The accuracy was within 5% and precision was <12% relative standard deviation. The method presented allows for analysis of 3-MP from biological samples for further drug developmental investigations of this promising cyanide antidote.
