TRPV1 antagonists have great potential for the treatment of pain. A case study on a drug candidate will be presented that will document the evolution of the chemical route through three stages of development. This will include detailed descriptions of the initial enabling route, innovative changes to the chemistry through route scouting and final demonstration of the optimized route at multi-kg scale.
