Asymmetric transfer hydrogenation (ATH) constitutes a powerful methodology for the synthesis of chiral secondary alcohols, giving products in high yield and enantioselectivity at low catalyst loadings. ATH has been successfully applied to a series of structurally related acetylenic ketones and aromatic ketones bearing a dioxin-4-one group. The fundamental principles of the reductions were established and detailed reaction conditions including the effects of hydrogen donors, catalyst loading, temperature and the stability of substrates were investigated. Chiral products from the ATH reactions serve as versatile and valuable building blocks and have also been used in natural product total synthesis.
