As part of our continuing program to identify interesting chemotypes for library development (for example, see: J. Comb. Chem. 2008 , 10, 628-631; Bioorg. Med. Chem. Lett. 2008 , 18, 4876-4879; ACS Comb. Sci. 2011 , 13, 443-448), we developed a new approach to polycyclic annulated indole scaffolds using a strategic combination of indole aryne cycloadditions (Org. Lett. 2007 , 9, 4135-4137; Tetrahedron Lett. 2009 , 50, 201-204; Org. Lett. 2009 , 11, 63-65; Tetrahedron Lett. 2009 , 50, 7113-7115; Org. Lett. 2010 , 12, 96-99) and cross-coupling reactions (e.g., Buchwald-Hartwig and Suzuki-Miyaura). In this presentation we describe our ongoing efforts to expand the utility of this tactic by introducing a ROM/RCM component to the overall process.
