The utility of poly(ethylene glycol) diacrylate (PEG-DA) hydrogels for osteochondral regeneration would be enhanced with spatially-controlled introduction of bioactivity and osteoinductivity. We have incorporated inorganic, hydrophobic methacrylated star polydimethylsiloxane (PDMSstar-MA) into PEG-DA-based hydrogels and have demonstrated that it induces the formation of hydroxyapatite (i.e. is bioactive) and promotes MSC lineage progression toward osteoblast-like fates (i.e. is osteoinductive) in proportion to its concentration within the scaffold.  The properties of the scaffolds (e.g. pore size, modulus, and PMDS distribution) were controlled with various fabrication parameters including wt% ratio of PEG:PDMSstar-MA, total wt% concentration of macromers in precursor solution and precursor solution solvent type (i.e. water or dichloromethane). Furthermore, scaffolds were prepared as continuous gradients for rapid screening of scaffold properties and also to prepare scaffolds ideally suited for generation of the osteochondral interface.
