Algae, a third generation biofuel feedstock, is a promising source for hydrocarbons. However, optimization of oil yield and reduction of process cost are essential for large-scale algal oil production.  Detailed knowledge of molecular composition can provide key information about cellular growth and guide subsequent production processing.  Nannochloropsis oculata was efficiently extracted by a combination of organic solvents.  Subsequent phase separation provides a polar lipid and a nonpolar lipid fraction. The polar lipid fraction was separated and detected by nano-LC LTQ 14.5 T FT-ICR MS. The nonpolar lipid fraction was analyzed by direct infusion FT-ICR MS. Structural assignment was based on accurate mass and tandem mass spectra. High mass resolution reveals isotopic fine structure to identify novel sulfate containing species. LC separation resolves structural isomers otherwise indistinguishable by MS alone.  This work was supported by NSF Division of Materials Research through DMR-0654118 and the State of Florida.
