Bioengineering of heterologous hosts to enable the production of commodity and pseudocommodity chemicals is an attractive alternative to current methods, which rely upon production from petroleum distillates.  Our team is pursuing metabolically engineered catechol and catechol-derived commodity chemicals, including cis, cis-muconic acid and the novel polymeric feedstock �-carboxy-cis,cis-muconic acid.  Both compounds are produced using a de novo biosynthetic pathway expressed from a multi-mono cistronic construct in an E. coli host.  Biosynthesis of this material is ultimately derived from shikimate pathway intermediates in the host strain.  Central to our process is the integration of a novel dehydroshikimate dehydrogenase (AsbF) identified in bacterial siderophore biosynthesis, which produces 3,4-dihydrobenzoic acid from dehydroshikimate.  Product isolation and characterization using TLC, NMR, and LC-MS methods will be discussed.  Biosynthetic production of this material provides a facile route to pseudocommodity chemicals with minimal chemical waste outputs relative to industrial processes.
