A chemically-defined medium was developed as part of a CHO cell culture platform process enabling speed-to-clinic for early clinical development projects. The final formulation of the chemically-defined medium included the most essential and generally beneficial media components based on data from multiple model cell lines. Those components not included in the platform were maintained in a �media component toolbox� for late stage bioprocess development. Data are presented from shake flask studies used to screen components for productivity enhancement. Hydrocortisone (cortisol), a glucocorticoid, was selected for further testing in a highly-controlled, 2L scale-down cell culture bioreactor model. Hydrocortisone, as a single supplement to the chemically-defined basal medium, was shown to increase specific productivity (productivity per cell) and result in an improved product quality profile relative to controls for responsive cell lines.
