DNA molecules can adopt various structures that are different from the canonical right-handed double-helix (B-DNA). DNA polymorphism is believed to contribute to various biological processes including genetic instability. Left-handed Z-DNA is stabilized in vivo by negative supercoiling, mutations, and by Z-DNA binding proteins. Circular dichroism is commonly used to identify DNA conformations. However, detection of one particular DNA form can be hampered by spectral overlaps. We will report the use of charged metalloporphyrins and conjugated metalloporphyrin dimers as highly selective and sensitive supramolecular probes for chiroptical detection of left-handed Z-DNA in the visible region of the electromagnetic spectrum. Detection of Z-DNA in the presence of excess B-DNA will also be presented.
