Coupling liquid separations to mass spectrometry has traditionally been done with spray ionization methods such as electrospray and atmospheric pressure chemical ionization. However, there are advantages to using lasers, either by themselves or together with liquid sprays, for coupling liquid chromatography or capillary electrophoresis separations to mass spectrometry. Lasers have the ability to remove material from a sample with minimal preparation under vacuum or ambient conditions. A wide variety of laser-based methods have been used to couple LC and CE to mass spectrometry, for example matrix-assisted laser desorption ionization can be used off-line, in continuous flow mode, or by desorption in a spray of aerosol particles. On-line laser desorption with electrospray ionization has recently been coupled to liquid separations with ambient sampling. An overview of the use of laser desorption for LC and CE-MS leading to the latest developments in ambient mass spectrometry will be presented.
