The effect of gold nanoshell-mediated photothermal heating on lymphatic endothelial cells (LECs) was investigated for potential application in photothermal therapy to disrupt tumor-associated lymphatics and inhibit lymphatic metastasis. Au nanoshells (AuNSs) are nanoparticles that absorb strongly in the near Infrared region and efficiently convert absorbed light to heat. Upon laser irradiation, a therapeutic dose of heat is generated which induces photothermal injury to LECs. The photothermal heating properties of AuNSs were characterized in vitro by measuring the temperature change upon laser irradiation. The effects of laser wavelength, laser power and AuNS concentration on the photothermal heating were examined. The efficacy of AuNS-mediated heating for inducing photothermal injury to LECs was investigated using the SV-LEC cell line. The uptake of AuNSs by LECs was studied. Cytotoxicity of AuNSs and the cell viability after laser irradiation was evaluated using LDH assay based on the measurement of lactate dyhydrogenase release from cells.
