In the present study, a range of potential oligomannan epitope sub-structures were systematically synthesized for further evaluation in lectin binding analyses. The sub-structures were subsequently applied to nanoparticle presentation, where a photoconjugation strategy was adopted. Underivatized carbohydrates were thus directly attached to fluorescently-doped silica nanoparticles, based on the photochemical properties of perfluorophenylazides, yielding strongly fluorescent, multivalent carbohydrate-presenting entities. The resulting epitope sub-structure-presenting nanoparticles were then applied to direct interrogation of different lectin microarrays: model arrays based on plan lectins, and arrays of cyanovirin-N lectins, benefitting from higher sensitivity owing to the multivalent nature of the interactions.
