There is increasing focus on biorenewables as an alternative to petroleum for the synthesis of chemicals and chemical precursors. We are introducing polyketide biosynthetic pathways to engineer Saccharomyces cerevisiae as a reliable source of renewable platform chemicals, including triacetic acid lactone (TAL). Our recent efforts have focused on the improvement of central carbon utilization towards the target compound via pathway modifications suggested by both computational modeling and relevant strategies found in the literature. These metabolic engineering efforts have allowed us to quickly reach greater than g/L titers experimentally using the wild-type Gerbera hybrida 2-pyrone synthase (2-PS). Novel synthase mutants were coupled with our engineered strains for further increases in productivity. Through a combination of expression condition optimization, metabolic pathway engineering, and enzyme engineering, we have increased TAL levels 25-fold.
