Trinitrobenzene sulfonic acid (TNBSA) method is used routinely for the quantitative estimation of lysine residues in any solution. However it has never been adapted to similar determinations in a heterogeneous system. In our laboratory, regenerated cellulose membranes were linked to lysine residues to prepare affinity membranes for protein isolation. Thereafter, to compare their performance with that to other separation matrices, it is critical to know the lysine density on membrane surface with high accuracy. The objective of this project was to modify the published TNBSA protocol to quantify the L-lysine bound to the membrane. Pieces of known dimensions were cut from the modified membranes and treated according to the Habib�s TNBSA method. The membrane pieces were then dissolved in concentrated HCl and the absorbance of resulting solution measured at 335 nm. Pure L-Lys was used as the standard solution while membranes consisting of Boc-Lys were used as negative controls.
