The real-time measurement of single nanoparticles in complicated system is significant for the materials and biosensing. In this work, we proposed a readily and high-throughput method to measure the size of single gold nanoparticles (GNPs) based on conventional dark-field microscopy (DFM). The localized plasmon resonance scattering light of GNPs could be captured by DFM. Then the RGB (three primary colors, red, green, and blue) chrominance information of the scattering light was converted into spectral wavelength and diameter of GNPs via matlab program according to the relevance between the sizes of GNPs and scattering light. Furthermore, this method supports real-time monitoring in versatile mediums including in liquid and living cells that is much more facile than scanning electron microscope (SEM). Therefore, our approach improved the ability of optical microscopy in a new way which could be widely applied in materials, sensors and cell imaging.
