Lateral flow assays (LFA; the 'pregnancy test' format) are simple and inexpensive diagnostics, highly suitable for point-of-care applications, but the sensitivities of the standard gold and latex particle formats are not as good as classical immunoassays.  Here, we introduce horseradish peroxidase labeled immuno-phage particles as reporters for enhanced-sensitivity LFAs.  The phage particles are engineered to express a small biotinylatable peptide that allows the convenient streptavidin-based attachment of any biotinylated affinity agent, including full-length antibodies without the need for cloning.  Sensitivities for model proteins with high-affinity antibodies are in the pM range, with phage concentration by PEG precipitation extending the limit of detection into the fM range.
